Overview

Intestinal mucosal proteins refer to a heterogeneous group of proteins that collectively maintain the structural and functional integrity of the gastrointestinal barrier. This group encompasses several distinct classes, including tight junction proteins (such as zonula occludens-1, claudins, and occludin) that regulate paracellular permeability, and mucins (such as MUC2 and MUC3) that constitute the protective mucus layer. Additionally, it includes antimicrobial peptides, secretory immunoglobulins (SIgA), and enzymes like intestinal alkaline phosphatase, which contribute to the chemical and immunological defense of the gut. These proteins are vital for preventing the translocation of luminal pathogens and toxins into the systemic circulation while facilitating nutrient absorption. Dysregulation or decreased expression of these proteins is a central feature of 'leaky gut' and chronic inflammatory conditions like inflammatory bowel disease (IBD). While not a single molecular target, the collective modulation of these proteins is a primary objective for therapeutic agents aimed at mucosal healing and barrier restoration.
