These folates act as essential cofactors in one-carbon transfer reactions involved in nucleotide biosynthesis, amino acid metabolism, and methylation cycles critical for gene regulation and cellular proliferation[2][1][3][5]. The pool's size and composition are determined by cellular import, export, metabolism, and retention processes, incorporating enzymes such as folylpolyglutamate synthetase (FPGS) and transporters, and is compartmentalized into cytosolic and mitochondrial pools. Disruption of folate homeostasis can have wide-ranging effects on DNA synthesis, repair, and epigenetic control, thereby impacting development, cancer, and cardiovascular disease risk[5][4][1][2][3].
