Overview

Intracellular metabolic machinery is a collective term for the integrated network of enzymes, cofactors, and organelles, such as mitochondria and the endoplasmic reticulum, that execute the biochemical reactions necessary for cellular life (StatPearls, 2023). This system encompasses fundamental pathways including glycolysis, the tricarboxylic acid (TCA) cycle, oxidative phosphorylation, and the biosynthesis of macromolecules like lipids and proteins (Nature Reviews Molecular Cell Biology, 2020). While not a single therapeutic target, the machinery is frequently reprogrammed in diseases such as cancer to support rapid growth, a phenomenon known as the Warburg effect (Cell, 2011). Hormones like insulin regulate this machinery by activating signaling cascades that shift the cell between anabolic and catabolic states (Guyton and Hall Textbook of Medical Physiology, 2020). Because these processes are ubiquitous and essential, pharmacological interventions must precisely target specific nodes within the machinery to avoid widespread systemic toxicity (Trends in Pharmacological Sciences, 2018).
