Overview

Intracellular protein–protein interactions (PPIs) are specific physical contacts established between proteins inside the cell that drive essential biological processes, such as cellular signaling, metabolism, gene expression, and the formation of supramolecular complexes[1][4]. These interactions can be transient or stable, homotypic or heterotypic, and are mediated by a range of molecular forces, including electrostatic interactions, hydrogen bonding, and hydrophobic effects[1][2][4]. Aberrant or dysregulated PPIs are implicated in diseases such as cancer, neurodegenerative disorders, and infections, making certain PPIs attractive, albeit challenging, targets for drug discovery[3][5]. However, "intracellular protein–protein interaction" is not a single, druggable molecule or receptor, but encompasses thousands of possible specific molecular interactions occurring in the intracellular environment.
