Overview

Intraflagellar transport protein 20 homolog (IFT20) is a small, highly conserved protein that is an integral component of the IFT complex B, which is essential for the assembly and maintenance of eukaryotic cilia and flagella[1][2][5]. IFT20 is unique among IFT proteins for its dual localization at both the cilium/basal body and the Golgi apparatus, acting as a dynamic adapter to facilitate trafficking of ciliary membrane proteins from the Golgi to the cilium. It is anchored to the Golgi via GMAP-210, and its function includes assembling IFT "trains" within the cilium, as well as mediating protein transport to the ciliary compartment[4][5]. IFT20 is implicated in cellular processes beyond ciliogenesis, notably in immune cell signaling (especially T cell activation) and regulation of receptor trafficking linked to cell migration and tumor invasiveness[4][5]. Defects in IFT20 are associated with a variety of ciliopathies and have roles in tumorigenesis and immune modulation, though it is not currently considered a direct drug target.
