IFT27 is a small Rab-like GTP-binding protein that forms a core part of the IFT-B complex required for the assembly and maintenance of cilia and flagella in eukaryotic cells[1][2][3][4]. It functions in both anterograde and retrograde intraflagellar transport by mediating the import and assembly of complex A and B proteins and is essential for cargo loading of retrograde transport machinery[1][2]. IFT27 acts together with IFT25, interacts directly with nucleotide-free ARL6 to help assemble the BBSome coat, and is required for proper ciliary signaling, including the removal of Patched-1 and Smoothened receptors during Hedgehog signaling[3]. Knockdown or mutation of IFT27 disrupts flagellar assembly, ciliary signaling, and can result in cell cycle and cytokinesis defects[2]. Although highly conserved, IFT27 is not present in certain model organisms whose ciliated cells are terminally differentiated[2].
