Overview

Intraflagellar transport protein 70A (IFT70A) is a core component of the intraflagellar transport (IFT) machinery, specifically acting as a subunit of the IFT-B complex, which is essential for the assembly and maintenance of cilia. IFT70A is a tetratricopeptide repeat (TPR)-containing protein required for the anterograde movement of protein cargos along the axonemal microtubules from the base to the tip of cilia, a process driven by kinesin-2 motors. It is also required for polyglutamylation of axonemal tubulin[2][6][8]. Mutations affecting IFT-B components, including IFT70A, can result in human genetic diseases known as ciliopathies, which impact development and organ function due to defective cilia structure and signaling[2][5]. IFT70A is characterized by its involvement in binding other IFT-B subunits, forming a tight spiral with a hydrophobic core, and maintaining the structural integrity of the IFT-B train critical for ciliary cargo transport[3][8]. Currently, IFT70A itself is not recognized as a direct therapeutic target (e.g., for small molecules or antibodies) and has no known clinically approved interacting drugs, mechanisms of action, or established biomarker applications. Its clinical relevance is mainly in the context of genetic diagnosis and research in ciliopathies[2][5].
