Overview

The iodine organification process is a key biochemical event in the thyroid gland during which inorganic iodide is oxidized and covalently linked to tyrosine residues of thyroglobulin, forming monoiodotyrosine (MIT) and diiodotyrosine (DIT), precursors to thyroid hormones T3 and T4. This process is catalyzed by the enzyme thyroid peroxidase and is critical for normal thyroid hormone biosynthesis. Defects or inhibition of organification—such as by antithyroid drugs like thionamides—can lead to thyroid disorders including hypothyroidism and goiter. Iodine organification is not a single molecular entity but rather a stepwise biochemical process essential for endocrine health.
