IQ motif and ubiquitin-like domain-containing protein (IQUB) is a multifunctional adapter/scaffold protein identified in humans, involved in regulating intracellular signaling and cell structure. It contains an amphiphilic IQ motif known for calmodulin binding and a ubiquitin-like domain, suggesting versatility in post-translational modification and protein-protein interactions. IQUB plays key roles in flagellated sperm motility (localizing to the radial spoke in the flagellum), and is predicted to be active in acrosomal vesicles and motile cilia. Pathologically, IQUB is upregulated in breast cancer and contributes to oncogenic signaling via Akt/GSK3β/β-catenin, enhancing cell proliferation and migration. Variants of IQUB have been associated with neurological disorders, including familial bipolar disorder, possibly through disruption of CREB and ERK/MAPK signaling important for neuronal and synaptic plasticity. IQUB also appears mutated in gastric cancer metastasis. Collectively, IQUB acts at the interface of structural cellular complexes and signal transduction, with dysregulation implicated in cancer progression and neuropsychiatric vulnerability.
