Overview

IQ motif containing GTPase activating protein 1 (IQGAP1) mRNA is the transcript encoding a versatile scaffold protein that integrates multiple intracellular signaling pathways, including the MAPK, PI3K/Akt, and Wnt/beta-catenin cascades (UniProt P46940). The encoded IQGAP1 protein plays a critical role in regulating the actin cytoskeleton, cell-cell adhesion, and cell migration by interacting with Rho GTPases like Cdc42 and Rac1 (PMID: 24608501). In many human malignancies, such as breast, lung, and liver cancer, IQGAP1 is significantly overexpressed and correlates with increased tumor aggressiveness, metastasis, and poor clinical outcomes (PMID: 30107118). Targeting the IQGAP1 mRNA using RNA interference (RNAi) or antisense oligonucleotides (ASOs) aims to deplete the protein levels, thereby disrupting the oncogenic signaling networks it supports (PMID: 28656238). Preclinical studies have demonstrated that silencing IQGAP1 mRNA can inhibit cancer cell proliferation, reduce invasion, and sensitize tumors to existing chemotherapies (PMID: 25533318). While currently in the preclinical stage, this approach represents a promising strategy for precision oncology and potentially for treating inflammatory conditions where IQGAP1-mediated signaling is dysregulated.

Mechanism of action

The primary mechanism of action for drugs targeting IQGAP1 mRNA is RNA interference (RNAi) or antisense-mediated degradation, which leads to the knockdown of IQGAP1 protein levels and subsequent disruption of oncogenic signaling scaffolds (PMID: 24608501, PMID: 28656238).
