Iron–sulfur cluster-containing proteins are a broad and essential class of metalloproteins characterized by the presence of iron–sulfur (Fe–S) clusters as prosthetic groups. These clusters, composed of iron and inorganic sulfide ions, are coordinated within the protein structure—typically via cysteine residues—and exist in several forms, most commonly [2Fe–2S], [3Fe–4S], and [4Fe–4S] clusters. The structural diversity allows these proteins to participate in a wide array of biological processes including electron transfer, catalysis, sensing cellular levels of iron or oxygen, DNA replication and repair, and biosynthesis pathways for cofactors. Some Fe-S clusters also serve purely structural roles.
