Iroquois-class homeodomain protein IRX-2 (IRX2) is a DNA-binding transcription factor encoded by the IRX2 gene, part of the Iroquois homeobox gene family[1][3][8]. This protein plays multiple roles in embryonic pattern formation, particularly during vertebrate development, by regulating the transcription of target genes through sequence-specific DNA binding[3][4][8]. IRX2 exerts negative regulation of gene expression, particularly through RNA polymerase II, and has been implicated in the regulation of cell proliferation, including modulation of MMP2 and VEGF expression in an AKT-dependent manner[4][7]. IRX2 is associated with disease states, particularly cancers such as kidney clear cell sarcoma and astrocytoma, and is progressively downregulated in HPV-positive cervical neoplastic keratinocytes, suggesting a role as a prognostic marker for cervical cancer progression[1][3][7]. There are currently no approved drugs targeting IRX2 directly, and its primary significance is as a developmental gene and disease biomarker rather than a direct therapeutic target[1][3][4][7][8].
