Overview

Iroquois homeobox protein 1 (IRX1) is a member of the Iroquois homeobox gene family, encoding a transcription factor with a distinctive homeodomain and IRO motif. IRX1 plays essential roles in early pattern formation during embryogenesis, regulating the development of organs such as the lungs, heart, eyes, and nervous system. In adult tissues, IRX1 is generally expressed at low levels, with higher expression in heart, adipose, kidney, and breast. IRX1 acts as a tumor suppressor in several cancer types, with its silencing by promoter hypermethylation contributing to malignancy (as in gastric and head and neck cancers). In some contexts, such as osteosarcoma, overexpression can contribute to metastatic potential. Because it is a nuclear transcription factor, IRX1 is not considered a classical druggable target at present. IRX1 is evolutionarily conserved and belongs to the homeobox protein superfamily, underlining its importance in developmental biology. Its dichotomous roles in cancer reflect context-dependent gene regulatory effects and epigenetic modulation, rather than direct ligand or drug targeting.
