Overview

ISL LIM homeobox 1 (ISL1) encodes a LIM/homeodomain transcription factor that binds to enhancer sequences, notably the insulin gene enhancer, and is centrally involved in the differentiation and development of several organ systems, including the pancreas, nervous system, and heart[3][4]. ISL1 is essential for the specification of motor neurons, plays a critical role in early cardiac progenitor cell lineages, and acts as a marker for cardiac and pancreatic progenitors through embryonic development[2][3][4]. It mediates gene regulatory networks influencing cell fate, proliferation, and organ formation, interacting with cofactors such as LDB1 and other LIM proteins, and participates in pathways like Jak/Stat signaling via complex formation with proteins such as Jak1 and Stat3[1]. Mutations and disruption of ISL1 are implicated in major developmental disorders, including cardiac anomalies and certain forms of diabetes[2][3][4]. Direct pharmacological targeting of ISL1 is not currently reported; its primary relevance is in developmental biology, disease modeling, and as a stem/progenitor cell marker.
