Iso-glutaminyl cyclase (QPCTL), also known as glutaminyl-peptide cyclotransferase-like protein or isoQC, is a zinc-dependent enzyme that catalyzes the intramolecular cyclization of N-terminal glutamine and glutamic acid residues into pyroglutamate (pGlu) at the N-terminus of proteins. This post-translational modification, called pyroglutamylation, influences protein stability and function. QPCTL plays a critical role in the maturation of chemokines and the CD47 "don't-eat-me" signal, impacting immune evasion by tumor cells. Targeting QPCTL offers therapeutic opportunities in cancer immunotherapy and inflammatory diseases.
