Overview

JADE1 (Jade family PHD finger protein 1, abbreviation JADE1, also known as PHF17) is a nuclear protein present in humans and other mammals that is characterized by two tandem PHD (Plant Homeo-Domain) zinc fingers—epigenetic histone code reader domains that bind histones and regulate chromatin structure. JADE1 associates with the histone acetyltransferase HBO1 to promote histone H4 acetylation, which enhances transcription and regulates DNA synthesis, cell cycle progression, and cytokinesis. The protein can function as a transcriptional co-activator and E3 ubiquitin ligase, and is implicated in cell apoptosis, differentiation, and suppression of tumor growth—particularly in renal tubular epithelial cells, where its expression and stability are linked to the tumor suppressor VHL protein. Reduced JADE1 activity impairs apoptosis, promotes cancer development, and disrupts cell proliferation control. JADE1 interacts with key cell cycle effectors and may be regulated by kinases including Aurora A/B. Current research positions JADE1 primarily as a chromatin regulator and tumor suppressor with emerging relevance for cancer therapy and cell proliferation control.
