Jade family PHD finger protein 3 (JADE3, also known as PHF16, KIAA0215, and PHD finger protein 16) is a nuclear protein characterized by tandem PHD (plant homeodomain) finger motifs that function in chromatin recognition and regulation[1][3][4]. As a member of the JADE protein family (JADE1, JADE2, JADE3), it acts as a core subunit and scaffold for some HBO1 (histone acetyltransferase binding to ORC1) complexes that mediate histone H4 acetylation, thereby influencing chromatin structure, gene expression, and transcriptional activation[1][3][4]. JADE family proteins are implicated in key biological processes including gene regulation, chromatin remodeling, and cellular differentiation. Although the detailed function of JADE3 specifically remains less well characterized than JADE1, it is considered a protein coding gene essential for chromatin modification and gene regulation. Dysregulation or mutation of JADE family members is linked to certain diseases, most notably cancer, underscoring their importance in maintaining genomic stability and cellular identity[1][3][4].
