JADE1 adjacent regulatory RNA (lncRNA-JADE)

Target
lncRNA-JADE
Molecular classification
Long non-coding RNA (lncRNA), Regulatory RNA, Other (not a protein, enzyme, receptor, or transporter; regulatory RNA acting mainly in epigenetic gene regulation)
01

Overview

JADE1 adjacent regulatory RNA, also known as lncRNA-JADE, is a highly conserved long non-coding RNA (~1.7 kb) transcribed adjacent to the JADE1 gene[1][4]. It is induced by DNA damage via the ATM/NF-κB axis and activates transcription of the neighboring JADE1 gene by recruiting the transcriptional co-activators BRCA1 and p300[1][3][4]. Physiologically, lncRNA-JADE supports cell proliferation and limits apoptosis, and is upregulated in breast cancer tissue compared to normal. Its transcriptional regulatory actions help coordinate chromatin acetylation and facilitate DNA damage recovery processes, making it biologically significant in cancer progression and potentially relevant for targeted therapeutic strategies[1][3][4].

Mechanism of action

Not directly drug-targetable, but acts by recruiting p300 and BRCA1 to the JADE1 promoter to promote transcription, influencing chromatin state and gene expression in the context of DNA damage response

03

Biological functions

Regulation of JADE1 gene transcriptionChromatin remodeling via recruitment of coactivator complexesDNA damage response (DDR), particularly ATM/NF-κB-dependent DNA damage signalingCell proliferation and apoptosis regulation; silencing leads to impaired proliferation and enhanced apoptosis, while overexpression has the opposite effect
04

Disease associations

Cancer (notably breast cancer, where it is upregulated and promotes tumorigenesis)Possible role in DNA repair deficiencies and related pathologies
05

Safety considerations

As a regulatory non-coding RNA, direct targeting presents challenges in specificity, off-target effects, and delivery for therapeutic modulation
06

Biomarkers

High lncRNA-JADE expression can serve as a biomarker for certain breast cancers and may correlate with proliferation indices and apoptosis resistance
