Overview

Janus kinase 1 and Janus kinase 2 are intracellular, non-receptor tyrosine kinases belonging to the Janus kinase family, which also includes JAK3 and TYK2. They associate with cytokine receptors and mediate intracellular signaling via the JAK-STAT pathway, which is critical for immune responses, hematopoiesis, and cell proliferation. JAK1 and JAK2 have non-redundant functions, with JAK2 being essential for erythropoiesis and hematopoietic cell development, while JAK1 is involved more broadly in cytokine signaling. Dysregulation or mutation of JAK2 (notably the V617F mutation) is a central pathogenetic event in various myeloproliferative neoplasms. Both kinases are important pharmaceutical targets, and several JAK inhibitors (jakinibs) have been developed for the treatment of immune-mediated diseases, certain cancers, and hematological malignancies. Their inhibition modulates immune function and cell growth but can be associated with immunosuppression and hematologic toxicities[1][2][3][4][5][6][7].
