Overview

Ruxolitinib is a small molecule kinase inhibitor primarily used to treat intermediate or high-risk myelofibrosis, polycythemia vera (especially in patients who are intolerant of or have an inadequate response to hydroxyurea), and both acute and chronic graft-versus-host disease (GVHD) after failure of prior therapies[1][3][4][5]. It is also approved as a topical cream for mild to moderate atopic dermatitis and non-segmental vitiligo[5]. Ruxolitinib acts as a potent, selective inhibitor of Janus kinase 1 (JAK1) and Janus kinase 2 (JAK2), which are tyrosine kinases involved in cytokine signaling pathways critical for hematopoiesis and immune function. By inhibiting JAK1/2, ruxolitinib blocks the dysregulated JAK-STAT pathway seen in myeloproliferative neoplasms, reducing abnormal blood cell proliferation, inflammation, splenomegaly, and related symptoms[4][7]. The drug was developed by Incyte.
