Overview

Janus kinase 1 and Janus kinase 2 are members of the Janus kinase (JAK) family, a group of intracellular, non-receptor tyrosine kinases that play a critical role in mediating cytokine signaling through the JAK-STAT pathway[1][2][3][4][5]. These proteins are associated with the cytoplasmic domains of various cytokine receptors and function by phosphorylating signal transducers and activators of transcription (STATs) once cytokines bind their receptors, thereby regulating gene expression relevant to immune function, blood cell production, and development[1][2][4][7]. Dysregulation of JAK1 or JAK2 is implicated in various pathologies, including inflammatory and autoimmune diseases, hematological malignancies, and myeloproliferative disorders[3][6]. Both JAK1 and JAK2 are therapeutically targeted by several approved kinase inhibitors, which modulate immune and hematopoietic pathways and are under active investigation for expanded indications[7][5][8].
