Overview

Janus kinase 2 (JAK2) with the V617F mutation is a constitutively active tyrosine kinase primarily involved in the pathogenesis of myeloproliferative neoplasms (MPNs). The V617F mutation disrupts the autoinhibitory function of the pseudokinase domain, leading to persistent activation of JAK2 and downstream STAT signaling, resulting in uncontrolled cell proliferation and survival in hematopoietic cells. Detection of this mutation is crucial for MPN diagnosis, and while targeted therapies exist, resistance remains a clinical challenge.
