Overview

The Janus kinase (JAK) family comprises four intracellular, non-receptor tyrosine kinases: JAK1, JAK2, JAK3, and TYK2. These enzymes are essential mediators of cytokine signaling and play a central role in the JAK-STAT pathway—a key mechanism for transducing signals from extracellular cytokines to the cell nucleus to regulate gene expression. They regulate immune responses, cell growth, proliferation, differentiation, and apoptosis. Dysregulation is implicated in various cancers and autoimmune diseases, making them important therapeutic targets.
