Overview

The Janus kinase pseudokinase domain (JH2) is a regulatory domain found in all members of the Janus kinase (JAK) family. The JH2 domain is located adjacent to the canonical tyrosine kinase domain (JH1) at the C-terminus of these proteins. It primarily functions as a negative regulator of the associated tyrosine kinase activity by direct interaction with the JH1 domain. Mutations in JH2 are implicated in hematological malignancies and other cancers, making it a potential therapeutic target.
