The JC polyomavirus Large T antigen (JC TAg) is a multifunctional viral protein crucial for the JCPyV life cycle. It plays a key role in viral DNA replication by binding to the viral origin of replication and recruiting host cell replication machinery. Additionally, JC TAg modulates host cell cycle pathways to create a cellular environment conducive to viral replication. It has oncogenic potential in some contexts. Targeting Large T antigen represents a potential therapeutic strategy for treating PML by disrupting viral replication.
