Overview

Jerky protein homolog (JRK) is a conserved nuclear protein structurally related to DNA-binding proteins such as the centromere-binding protein B (CENPB)[3][11]. It contains a helix-turn-helix DNA-binding motif and may act as a transcription factor or gene regulator. Disruption of the related gene in mice leads to recurrent seizures similar to human idiopathic generalized epilepsy, and the human gene is considered a candidate locus for epilepsy such as childhood absence epilepsy[2][3]. JRK is also implicated in tumorigenesis by promoting Wnt pathway hyperactivation and is reportedly overexpressed in various cancer types[1]. Alternative splicing produces multiple transcript variants. There is no current evidence of drugs directly targeting JRK or use as a clinical biomarker, and safety concerns specific to therapeutic manipulation of this protein are not documented in the literature.
