Overview

Joint stiffness is a clinical symptom characterized by the sensation of difficulty moving a joint or a restricted range of motion, often occurring after periods of inactivity or upon waking. It is not a discrete molecular target such as a protein or receptor, but rather a phenotypic manifestation of various underlying pathological conditions, most notably inflammatory arthritides and degenerative joint diseases. In inflammatory conditions like rheumatoid arthritis, stiffness is driven by the accumulation of inflammatory cytokines (e.g., TNF, IL-6) and synovial fluid within the joint capsule. Conversely, in osteoarthritis, it results from mechanical wear, loss of cartilage, and subchondral bone changes. While drugs like NSAIDs and biologics are used to alleviate joint stiffness, they do so by targeting specific molecular mediators of the primary disease rather than 'stiffness' itself. Consequently, this entry is classified as incorrect for a molecular target identification task.
