Jumonji domain-containing histone demethylases (JmjC-KDMs) are a family of enzymes that catalyze the removal of methyl groups from lysine residues on histone proteins. This demethylation process is crucial for regulating chromatin structure and, consequently, gene expression. The activity of these enzymes requires Fe(2+) and α-ketoglutarate as cofactors. Alterations in Jumonji domain-containing demethylases have been implicated in various human cancers due to their role in epigenetic regulation. As such, they are considered emerging therapeutic targets for anticancer drug development.
