Jumonji domain-containing protein 4 (JMJD4) is a member of the JmjC-domain-containing family of 2-oxoglutarate- and iron-dependent oxygenases[2][4][6]. It specifically functions as a C4 lysyl hydroxylase, catalyzing posttranslational hydroxylation of the eukaryotic release factor 1 (eRF1) at lysine 63, a modification that is essential for optimal translational termination and fidelity of protein synthesis[2][6]. JMJD4 is found in the cytoplasm and plays a crucial role in ensuring stop codon recognition during translation, acting independently of chromatin modification (unlike some JmjC proteins)[2][4]. While its core function is linked to fundamental processes of genome expression and protein quality control, direct links with specific human diseases remain only sparsely described, with some nominal association to rare tumors[6].
