Overview

Jumonji domain-containing protein 8 (JMJD8) is a small, endoplasmic reticulum luminal protein unique among the JmjC domain family, lacking the canonical amino acid motif required for classical histone demethylase activity[1][2][3]. It plays roles in protein complex assembly, protein folding, and is a positive regulator of TNF-induced NF-κB signaling, supporting inflammation and cellular response pathways[3][4]. JMJD8 interacts with metabolic proteins such as pyruvate kinase M2, is implicated in regulating angiogenesis and metabolism, and has documented oncogene-like activity in several cancers[5]. Recently, it was proposed as a biomarker for M2 macrophages and is associated with neurodegenerative disorders including Spinocerebellar Ataxia[4][5]. Its precise molecular functions remain under investigation, but emerging evidence positions JMJD8 as a molecular nexus in inflammation, ER homeostasis, and possibly metabolic pathologies. There are currently no reported drugs or selective inhibitors/activators, nor characterized direct mechanisms of pharmacological action. Overexpression or dysregulation has potential safety concerns due to ER stress and oncogenic potential[3][5].
