Overview
Junctional adhesion molecule 2 (JAM2) is a cell surface protein of the immunoglobulin superfamily, primarily located at tight junctions of endothelial and epithelial cells. JAM2 mediates cell-cell adhesion through homophilic and heterophilic interactions, notably with JAM3 and integrins (particularly α4β1), controlling paracellular permeability, leukocyte transmigration, cell polarity, and vascular integrity. It is critical in immune responses, stem cell homing, spermatogenesis, and myogenesis. Alterations in JAM2 are associated with various pathologies, such as cancer, inflammatory and autoimmune diseases, and vascular disorders. JAM2 is used as a prognostic biomarker in certain cancers and can be detected in tissue and plasma via immunoassays. Therapeutic targeting is challenging due to its essential physiological roles and wide tissue distribution[1][2][4][5][7].
