Overview

Jupiter microtubule-associated homolog 1 (JPT1) is a multifunctional cytosolic and microtubule-associated protein implicated in the regulation of cell cycle, cell adhesion, and multiple oncogenic signaling pathways[1][2][4]. It modulates AKT-mediated GSK3B signaling, regulates β-catenin (CTNNB1) stability, and influences androgen receptor (AR) signaling via proteasomal degradation of the receptor[2][4]. JPT1 is upregulated in several cancer types and is associated with increased tumor proliferation and progression[1]. In endometrial cancer, JPT1 serves as a predictive and pharmacodynamic biomarker for metformin response, with higher pre-treatment levels in responders and significant post-treatment decreases[1][3]. Elevated JPT1 is also linked to aggressive abilities in prostate and gastric cancer cell lines, mediated via competitive endogenous RNA (ceRNA) networks that enhance its expression[1]. Therefore, JPT1 functions not only as a biomarker but also as an effector in tumor biology, although it is not presently a direct drug target.

Mechanism of action

For metformin: Changes in JPT1 abundance serve as a pharmacodynamic marker of metformin response in endometrial cancer, but JPT1 is not a direct target of the drug.
