Kallikrein-related peptidase 5 (KLK5) is a serine protease primarily expressed in the epidermis and plays a crucial role in skin barrier formation and desquamation. It processes profilaggrin to filaggrin, an essential protein for epidermal differentiation and barrier integrity. KLK5 can activate other kallikreins, initiating a proteolytic cascade important for tissue homeostasis and repair. Dysregulation of KLK5 is associated with skin diseases like Netherton syndrome and has potential as a biomarker for ovarian cancer.
