Overview

KASH domain-containing protein 5 (KASH5) is a cytoskeletal adaptor protein that is a component of the LINC (Linker of Nucleoskeleton and Cytoskeleton) complex, which connects the nuclear envelope to the cytoskeleton to enable mechanical force transmission and nuclear positioning. KASH5 is essential for proper telomere attachment to the nuclear envelope during meiotic prophase, critical for homologous chromosome pairing and segregation in spermatocytes and oocytes. It recruits cytoplasmic dynein to telomere attachment sites at the nuclear envelope, a key step in chromosome movement during meiosis. Mutations in KASH5 are implicated in reproductive disorders such as premature ovarian failure and spermatogenic failure[1][2]. It is considered a structural and scaffolding adaptor rather than a classic therapeutic target like a receptor, enzyme, or transporter. No drugs are currently known to interact with KASH5, and it is not part of standard biomarker panels or known for specific safety concerns[1][2][3].
