Overview

KAT8 regulatory NSL complex subunit 1-like protein (KANSL1L) is a nuclear protein encoded by the KANSL1L gene in humans and is predicted to be a non-catalytic component of the NSL (nonspecific lethal) histone acetyltransferase complex[5]. It is thought to function as a scaffolding subunit within this chromatin regulatory assembly, enabling histone acetyltransferase activity through binding interactions, and thus participates in the regulation of transcription via chromatin modification[5][7]. KANSL1L is highly related to its paralog KANSL1, which acts as a core scaffold in the NSL complex and is implicated in enhancer regulation, cell proliferation, and mitosis[2][5]. While there is no direct evidence that KANSL1L itself is a therapeutic target, nor are there known drugs or clinical biomarkers, alterations in related NSL complex subunits are associated with cancer and neurodevelopmental disorders[2][3][5]. The specific biological and pathophysiological roles of KANSL1L remain less thoroughly characterized, but the protein is believed to support histone acetylation-related functions crucial for transcriptional regulation and possibly chromatin organization[5][7].
