Overview

KAT8 regulatory NSL complex subunit 3 (KANSL3) is a non-catalytic component of the NSL (Non-Specific Lethal) histone acetyltransferase complex, which plays a key role in mediating acetylation of histone H4 at lysine 5 and lysine 8 at transcription start sites, promoting transcription initiation and regulating gene expression in both the nucleus and mitochondria[2][4][6]. KANSL3 is required for the proper association of the catalytic subunit KAT8 with mitochondrial DNA and is also necessary for the transcription of genes involved in intraciliary transport, essential for cilium assembly and microtubule organization in both ciliated and non-ciliated cells[2][4][6]. Disruption of KANSL3 can lead to defects in cell structure and development (notably in kidney podocytes and mouse embryonic development) and is associated with developmental disorders such as Koolen-De Vries syndrome and liver fibroma[2][4]. KANSL3 is not known to be directly targeted by drugs and is not a therapeutic target (e.g., receptor, enzyme, transporter) but instead functions as an essential regulatory subunit within a chromatin-modifying complex[2][4][6].
