Overview

Katanin p80 WD40 repeat-containing subunit B1 (KATNB1) is the regulatory subunit of the microtubule-severing katanin complex, a heterodimer that consists of a catalytic p60 subunit (AAA ATPase KATNA1) and the p80 regulatory subunit encoded by KATNB1[1][3][5]. The p80 subunit directs the catalytic activity of katanin to centrosomes and other cellular locations and fine-tunes microtubule severing. KATNB1 orchestrates cytoskeletal remodeling during mitosis, regulates centriole and cilia numbers, and is essential for processes such as spindle formation, cytokinesis, and normal neurogenesis and sperm development[1][2][3]. Mutations in KATNB1 disrupt these processes, leading to severe neurodevelopmental defects (microlissencephaly, microcephaly), male infertility, and impaired ciliary signaling. The gene is highly evolutionarily conserved, highlighting its critical role in cell division and differentiation[3]. There are currently no known drugs targeting KATNB1 directly or clinically validated mechanisms of action or biomarkers based on its modulation[1][3].
