KCNJ8 antisense RNA 1 (KCNJ8-AS1)

Target
KCNJ8-AS1
Molecular classification
lncRNA, antisense RNA, non-coding RNA
01

Overview

KCNJ8 antisense RNA 1 (KCNJ8-AS1) is an antisense long non-coding RNA gene located in the human genome. It is transcribed from the opposite DNA strand of the KCNJ8 locus. KCNJ8-AS1 belongs to the class of lncRNAs, a group of regulatory RNA molecules exceeding 200 nucleotides in length, and is thought to regulate the expression of its sense gene, KCNJ8 (Kir6.1), which encodes an ATP-sensitive potassium channel involved in the regulation of vascular tone, cardiac electrophysiology, and linked to cardiac disorders such as cardiomyopathy and atrial fibrillation. The major biological role of KCNJ8-AS1 is gene expression regulation, possibly through modulation of KCNJ8 mRNA transcription, stability, or epigenetic status, as described for antisense lncRNAs in the literature. KCNJ8-AS1 is not a protein-coding gene nor a classical therapeutic target such as a receptor or enzyme, and no drugs are currently known to interact with it directly.

02

Mechanism of action

Not applicable. lncRNAs regulate gene expression epigenetically or via RNA-RNA interactions rather than by direct binding of small molecules or drugs.

03

Biological functions

Regulation of gene expression (specifically, regulation of KCNJ8/Kir6.1 and potentially its mRNA stability or transcription)
04

Disease associations

CardiomyopathyDilated cardiomyopathy (type 1O)Atrial fibrillation, familial (type 12)
05

Safety considerations

Not applicable as a therapeutic target; challenges are related to the general difficulty in targeting lncRNAs due to their lack of conserved structure, sequence, or ubiquitous expression patterns
06

Biomarkers

No established use as a clinical biomarker, but associations with cardiac diseases may suggest potential as a research biomarker for heart diseases
