KCNMA1 antisense RNA 1 (KCNMA1-AS1) is a long non-coding RNA (lncRNA) found in human cells that primarily resides in the nucleus and plays a critical role in the osteogenic differentiation of human bone marrow mesenchymal stem cells (hBMSCs)[4][6]. KCNMA1-AS1 is upregulated during osteogenic differentiation and directly interacts with the protein SMAD9, activating the SMAD9 pathway to promote bone formation both in vitro and in vivo[4][6]. Knockdown of KCNMA1-AS1 leads to reduced osteogenic differentiation, while its overexpression enhances bone formation. Preclinical research suggests KCNMA1-AS1 is a candidate biomarker and potential therapeutic target in bone defect and regeneration[4][6]. There is no evidence of direct pharmacological targeting; however, modulating this lncRNA or its interaction with SMAD9 represents a novel approach for bone tissue engineering and regenerative medicine. There are currently no known drug interactions and safety/efficacy profiles remain uncharacterized in clinical settings.
