Overview

Kelch domain-containing protein 7B (KLHDC7B) is a 594–595 amino acid protein featuring a characteristic Kelch domain in its C-terminal region, involved in protein-protein interactions[1][3]. KLHDC7B has been implicated in cancer biology, most notably in breast cancer, where its upregulation correlates with higher tumor grade, increased cell proliferation (as reflected in higher Ki67 indices), and poor differentiation[1][2]. Its expression is variable between tumor subtypes but generally higher in more aggressive and poorly differentiated tumors, such as grade 3 ductal carcinomas[1]. KLHDC7B’s functional role remains partially understood, but gene regulation studies in breast cancer cell lines indicate involvement in regulating apoptosis, proliferation, cell migration, and associations with long non-coding RNAs like STAR1[3]. Its upregulation in cancer has made it a candidate biomarker for prognosis and possibly future therapeutic targeting, but no direct modulators or drugs are known[1][3][5]. KLHDC7B is considered a member of the Kelch-like superfamily but does not fit neatly into classical categories such as receptor or enzyme[4].
