Kelch-like family member 34 (KLHL34) is a protein encoded by the KLHL34 gene and belongs to the Kelch-like (KLHL) gene family[3][5][7]. Like other KLHL proteins, KLHL34 contains a BTB/POZ domain, a BACK domain, and 5–6 Kelch motifs. Proteins of this family act as substrate-specific adaptors for Cullin-3-containing E3 ubiquitin ligase complexes, recruiting target proteins for ubiquitination and subsequent proteasomal degradation[4][5][6]. While the general function of the KLHL family involves diverse cellular processes such as protein homeostasis and cell signaling, the specific biological roles and substrates of KLHL34 have not been well defined in the literature[5][6][7]. There are reports that the broader KLHL family is associated with Mendelian diseases, muscle development disorders, and some cancers, but there is currently no direct evidence linking KLHL34 itself to therapeutic targeting, drug interaction, or use as a clinical biomarker[6]. Therefore, KLHL34 is not considered a validated therapeutic target or disease biomarker at this time.
