Overview

Kelch-like protein 21 (KLHL21) is a member of the Kelch-like protein family that functions primarily as a substrate-specific adapter for the Cullin 3-based E3 ubiquitin-protein ligase complex, contributing to protein ubiquitination and the regulation of chromosome passenger complex localization at the spindle midzone during mitosis[3][1]. KLHL21 is essential for cytokinesis and correct cell division, as well as negatively regulating the activation of IκB kinase-β (IKKβ), thereby modulating NF-κB signaling[1]. Dysregulated expression of KLHL21 promotes cancer cell proliferation, migration, and invasion, particularly in hepatocellular carcinoma, where it may serve as a prognostic biomarker and potential therapeutic target[2][3].
