Overview

Kelch-like protein 22 is a BTB-Kelch family adaptor protein that forms a functional cullin 3-RING E3 ubiquitin ligase complex (BTB-CUL3-RBX1). It mediates ubiquitin-dependent regulation of target proteins, notably monoubiquitination of Polo-like kinase 1 (PLK1), critical for regulation of spindle assembly checkpoint and proper chromosome segregation during mitosis. KLHL22 also regulates proteasomal degradation of DEPDC5, leading to mTORC1 pathway activation, thereby influencing cell growth, autophagy, and potentially tumorigenesis and cellular aging. KLHL22 overexpression correlates with poor prognosis in triple-negative breast cancer, and its expression serves as an independent prognostic marker for disease progression and survival. KLHL22 is primarily localized to the cytoplasm and is involved in the regulation of the spindle assembly checkpoint and cellular response to nutrient cues, affecting broad biological processes across different tissues.
