Kelch-like protein 27 (KLHL27), also known as intracisternal A particle-promoted polypeptide (IPP), is a member of the Kelch-like (KLHL) protein family, which collectively function as substrate adaptors for cullin 3 (CUL3)-based E3 ubiquitin ligase complexes[1][2][4][5]. Like other KLHL proteins, KLHL27 contains BTB/POZ, BACK, and Kelch domains that mediate protein–protein interactions, particularly linking substrates (such as actin) to the ubiquitination machinery for their modification or degradation[1][2]. KLHL27 is implicated in actin cytoskeleton organization and is highly expressed in some cancers, including breast cancer[2][4]. In cancer, potential interactions with proteins such as Hook2 (important for mitosis and cytokinesis) and Lig4 (involved in DNA repair) suggest contextual relevance, but no direct evidence currently supports its use as a therapeutic target or interaction with approved drugs[2][4]. **Note:** There is no evidence that KLHL27/IPP is a receptor, enzyme, transporter, or target of any approved or investigational drugs at this time. It is not currently considered a therapeutic target.
