Overview

Kelch-like protein 33 (KLHL33) is a member of the kelch-like (KLHL) protein family, characterized by the presence of BTB/POZ, BACK, and multiple Kelch domains[2][5][10]. KLHL33 encodes a substrate adapter protein that is thought to interact with Cullin3 to form part of the Cullin-E3 ubiquitin ligase complex, facilitating the ubiquitination and subsequent proteasomal degradation of specific substrate proteins[5]. This function is important for regulating protein stability and is implicated in processes such as cellular differentiation, maintenance of homeostasis, cell signaling, and response to oxidative stress[2][5]. KLHL33 is a protein-coding gene and shows tissue-specific expression, particularly enriched in skeletal muscle and brain tissues[2][8]. Disorders associated with variations in the KLHL33 gene include meningeal sarcoma and Takayasu arteritis, although the mechanistic link is not fully defined and KLHL33 itself is not currently a validated therapeutic target[7]. Most research on KLHL family proteins has focused on other members, such as KLHL40, KLHL41, and KEAP1, with the functional roles of KLHL33 remaining largely unexplored[5]. There are no known interacting drugs, reported biomarker uses, or established safety concerns for KLHL33 at this time.
