Overview

Kelch-like protein 35 (KLHL35) is a member of the large Kelch-like (KLHL) protein family, characterized structurally by the presence of a BTB/POZ domain, a BACK domain, and multiple Kelch repeats, typically forming a β-propeller structure. KLHL35 is predicted to function as a ubiquitin-like ligase-substrate adaptor, participating in the proteasome-mediated, ubiquitin-dependent degradation of cellular proteins via its role in the Cul3-RING ubiquitin ligase complex. While its exact physiological functions remain incompletely understood, KLHL35 is implicated in regulating cell growth, possibly acting as a tumor suppressor. Epigenetic silencing via promoter hypermethylation is associated with several cancer types, including renal cell carcinoma and hepatocellular carcinoma, indicating roles as both a potential tumor suppressor and a disease biomarker. KLHL35 expression is also reportedly affected by exposure to certain viral infections (e.g., HPV, HBV) and those changes may serve as biomarkers for disease risk or progression. Despite its relevance in cell growth regulation and disease association, KLHL35 is not a known target of therapeutic drugs or a classic “receptor,” “enzyme,” or other major druggable protein class.
