Overview

Kelch-like protein 5 (KLHL5) is a member of the kelch superfamily, characterized by BTB, BACK, and kelch repeat domains, which enable it to function as a substrate adaptor in Cullin-3 (CUL3)-based E3 ubiquitin ligase complexes. KLHL5 binding targets specific proteins for ubiquitination and degradation and is involved in regulating the actin cytoskeleton, cell morphology, and cell cycle progression. It is broadly expressed across benign and malignant tissues, with dysregulated expression in multiple cancer types; higher expression is often associated with poor prognosis, especially in gastric cancer, where it additionally modulates immune cell infiltration. KLHL5 knockdown in vitro increases cellular sensitivity to a variety of anticancer drugs, indicating potential as a therapeutic target or modulator of drug response. Its molecular function and interactors beyond CUL3 complexes remain incompletely characterized.
