Kelch repeat and BTB (POZ) domain-containing protein 8 (BTBD8) is a highly conserved protein found in both mouse and human. It contains a BTB/POZ domain, a BACK domain, and five kelch repeats, classifying it within the BTB-kelch (BBK) protein family[1][4]. The BTB domain is primarily implicated in mediating protein-protein interactions including dimerization and oligomerization, while the kelch repeats often relate to cytoskeletal association[1][2]. BTBD8 is ubiquitously expressed, with notable localization to the Golgi apparatus in non-dividing cells and translocation to the spindle apparatus during mitosis, suggesting roles in cell proliferation[1][4]. In neurons, it is involved in synaptic vesicle budding, endocytosis, and recycling, processes required for normal synaptic transmission[3][6]. Currently, no drugs are known to specifically target BTBD8, and it is not established as a classical therapeutic target[3][6]. Genetic studies have associated mutations in BTBD8 with certain immunodeficiency conditions[3][5].
