Overview

Kelch repeat and BTB domain-containing protein 11 (KBTBD11) is a multifunctional regulatory protein containing both a BTB/POZ domain and Kelch repeats[5][1]. It is known to participate in the regulation of cell signaling, cell proliferation, and differentiation. In bone, KBTBD11 acts as a negative regulator of osteoclastogenesis by interacting with the E3 ubiquitin ligase Cullin3, promoting the ubiquitination and proteasomal degradation of NFATc1, a key transcription factor in osteoclast differentiation[2][3][5]. KBTBD11 also functions in adipocyte biology, where its expression is nutritionally regulated and is required for adipocyte differentiation and triglyceride accumulation[4][5]. In cancer, especially colorectal carcinoma, polymorphisms in KBTBD11 have been linked to disease susceptibility and tumor suppressor function; reduced KBTBD11 expression enhances cell proliferation and impairs apoptosis[5]. KBTBD11 gene methylation patterns serve as biomarkers in oncology, including in acute lymphoblastic leukemia[5]. Currently, there are no known therapeutic drugs directly targeting KBTBD11, and it is not considered a classic therapeutic target such as a receptor or enzyme[5].
