Kelch repeat and BTB domain-containing protein 6 (KBTBD6) is a cytoplasmic and nuclear protein, encoded on human chromosome 13q14.11, that functions as a substrate-specific adaptor for the Cullin-3 RING-type E3 ubiquitin ligase complex (CUL3)[1][5]. KBTBD6 operates primarily as part of the CUL3(KBTBD6/7) E3 ubiquitin ligase complex, recruiting the guanine exchange factor TIAM1 for RAC1 and mediating its Lys-48–linked ubiquitination and subsequent proteasomal degradation[2][4][3]. Regulation of TIAM1 abundance by KBTBD6 controls RAC1 GTPase signaling, affecting actin cytoskeleton organization, cell migration, and proliferation. KBTBD6 possesses an ATG8 interaction motif (AIM) for interactions with GABARAP, and Kelch repeats for binding TIAM1; the BTB domain enables interaction with CUL3[3][4]. No direct links to therapeutic drugs, well-validated disease biomarkers, or major safety concerns have been established for KBTBD6 in the current literature.
